Radek Šachl: Unraveling Nanoscale Membrane Organization

Recent biophysical experiments have revealed the intricate and non-homogeneous distribution of lipids and proteins within cell membranes at the nanoscale, where spontaneous lipid aggregation and protein oligomerization give rise to larger functional units crucial for the proper functioning of biological membranes. However, characterizing these nanoscopic structures under native or near-native conditions remains a formidable technological challenge. In my talk, I will introduce two innovative in-house developed fluorescence microscopy techniques that provide unprecedented insights into the nanoscopic organization of membranes: Monte-Carlo-FRET offering nanometer resolution in xyz and dual(+1)-FCS (fluorescence correlation spectroscopy) that goes beyond simple visualization by enabling discrimination between functional and non-functional membrane-associated protein oligomers.
